Gut-associated lymphoid tissues (GALTs) comprise key intestinal immune inductive sites, including the Peyer’s patches of small intestine and different types isolated follicle (ILF) found along length gut. Our understanding human GALT is limited due to a lack protocols for their isolation. Here we describe technique that, uniquely among cell isolation protocols, allows identification all GALT, as well GALT-free lamina propria (LP). The involves mechanical separation mucosa from submucosa, allowing submucosal ILF (SM-ILF), LP-embedded mucosal (M-ILF) LP free contaminating tissue.
